Adam Armstrong scored the only goal of the game in the second half as Southampton overcame a sub-par Leeds side.
The win means Southampton will return to the Premier League after a one-year absence, while Leeds will remain in the Championship for another season.
Southampton have now won 29 matches across all competitions this season, their most since their promotion to the Premier League in 2012.
Leeds, meanwhile, have now lost three of their last four matches, and they will be disappointed not to have won promotion after finishing the regular season in second place.
